Transcripts with alternative 5′-untranslated regions (UTRs) result from the activity of alternative promoters and they can determine gene expression by influencing its stability and translational efficiency, thus executing complex regulation of developmental, physiological and pathological processes.

SHP-1, a protein-tyrosine phosphatase with two srchomology 2 domains, is expressed predominantly in hematopoietic and epithelial cells and has been implicated in numerous signaling pathways as a negative regulator. Two promoters direct the expression of human and murine SHP-1, and two types of transcripts (I) and (II) SHP-1, are initiated from each of these promoters. The cDNA sequences of (I)SHP-1 and (II)SHP-1 are identical except in the 5¢ untranslated region and in the first few coding nucleotides.

The intrachloroplastic localization of post-transcriptional gene expression steps represents one key determinant for the regulation of chloroplast development. We have character-ized an RNA binding protein of 63kDa (RBP63) from Chlamydomonas reinhardtiichloroplasts, which cofraction-ates with stromal thylakoid membranes. Solubility proper-ties suggest that RBP63is a peripheral membrane protein.

Interleukin (IL)-10 was cloned from the common carp (Cyprinus carpioL.) using IL-10 primers from carp head kidney following stimulation with concanavalin A and lipopolysaccharide. The cDNA consisted of a 1096 bp se-quence containing a 55 bp 5¢ untranslated region and a 498 bp 3¢ untranslated region. An open reading frame of 543 bp encoded a putative 180 amino acid protein with a putative signal peptide of 22 amino acids.

Interleukin (IL)-11 is a multifunctional cytokine that stimulates hematopoi-etic progenitor cells and exerts a series of important immunomodulatory effects. It was believed to be restricted to mammals, but here we report the first nonmammalianIL-11 gene, in rainbow trout (Oncorhynchus mykiss). A trout IL-11 cDNA clone was isolated that contains a 5¢-untranslated region (UTR) of 400 bp, an open reading frame of 612 bp and a large 3¢-UTR of 1924 bp.

Chronic hepatitis C virus (HCV) infection is a clinically important liver disease with limited therapeutic options in a significant proportion of patients. Therefore, novel efficient therapeutic agents are needed. Because the 5¢- and 3¢-untranslated regions (UTRs) of HCV are highly conserved and functionally important for HCV replication, they are attractive targets for RNA-cleaving ribozymes or small interfering RNAs (siRNAs).

Recognition of viral nucleic acids by vertebrate host cells results in the syn-thesis and secretion of type I interferons (IFN-a⁄b), which induce an anti-viral state in neighboring cells. We have cloned the genes and promoters of two type I IFNs from Atlantic salmon. Both genes have the potential to encode IFN transcripts with either a short or a long 5¢-untranslated region, apparently controlled by two distinct promoter regions, PR-I and PR-II, respectively.
