This study reports the identification and characterization of the regulatory subunit, TbRSU, of protein kinase A of the parasitic protozoon Trypanosoma brucei. TbRSU is coded for by a single copy gene. The protein contains an unusually long N-terminal domain, the pseudosubstrate site involved in binding and inactivation of the catalytic subunit, and two C-terminally located, closely spaced cyclic nucleotide binding domains.

NTPDase1 and NTPDase2 are two related plasma mem-brane-located enzymes involved in the extracellular degra-dation of nucleoside 5¢-tri- and -diphosphates. They differ regarding their hydrolysis ratios for ATP and ADP. The binding of several alkyl- and aryl-guanidines andN-hydroxyguanidines to the oxygenase domain of inducible NO-synthase (iNOSoxy) was studied by UV⁄Vis difference spectroscopy. In a very general manner, monosubsti-tuted guanidines exhibited affinities for iNOSoxy that were very close to those of the corresponding N-hydroxyguanidines. Two structures of monomeric methionyl-tRNA synthetase, fromMycobac-terium smegmatis, in complex with the ligands methionine⁄adenosine and methionine, were analyzed by X-ray crystallography at 2.3 A˚ and at 2.8 A˚ , respectively. The structures demonstrated the flexibility of the multidomain enzyme. A new conformation of the structure was identified in which the connective peptide domain bound more closely to the catalytic domain than described previously.
