Regulator of G protein signaling (RGS) proteins are gatekeepers regulating the cellular responses induced by G protein-coupled receptor (GPCR)-mediated activation of heterotrimeric G proteins. Specifically, RGS proteins determine the magnitude and duration of GPCR signaling by acting as a GTPase-activating protein for Gα subunits, an activity facilitated by their semiconserved RGS domain. The R7 subfamily of RGS proteins is distinguished by two unique domains, DEP/DHEX and GGL, which mediate membrane targeting and stability of these proteins.

Metabolically unstable proteins are involved in a multitude of regulatory networks, including those that control cell signaling, the cell cycle and in many responses to physiological stress. In the present study, we have deter-mined the stability and characterized the degradation process of some members of the Gq class of heterotrimeric G proteins. Pulse-chase experi-ments in HEK293 cells indicated a rapid turnover of endogenously expressed Gaq and overexpressed Gaq and Ga16 subunits.

Replication protein A (RPA), a heterotrimeric protein composed of 70, 32 and 14-kDa subunits, has been shown to be essential for DNA replication, repair, recombination, and transcription. Previously, we found that, in two seed plants, rice and Arabidopsis, there are two different types of RPA70-kDa subunit. Substantial biochemical and genetic characterization of these two subunits, termed OsRPA70aand OsRPA70bor AtRPA70a and AtRPA70b, respectively, is described in this report.

Transport of presecretory proteins into the mammalian rough endoplasmic reticulum involves a protein translocase that comprises the integral membrane proteins Sec61ap, Sec61bp, and Sec61cp as core components. Electron microscopic analysis of protein translocase in rough micro-somal membranes suggested that between three and four heterotrimeric Sec61p complexes form the central unit of protein translocase.

In this study, its three subunits were cloned and sequenced from the venom gland cDNAs of Daboia siamensis. We have investigated the existence of a precoupled form of the distal C-ter-minal truncated cannabinoid receptor 1 (CB1-417) and heterotrimeric G proteins in a heterologous insect cell expression system. CB1-417 showed higher production levels than the full-length receptor. The production lev-els obtained in our expression system were double the values reported in the literature.
