  • Although the microbiome is established as an important regulator of health and disease, the role of viruses that inhabit asymptomatic humans (collectively, the virome) is less defined. While we are still characterizing what constitutes a healthy or diseased virome, an exciting next step is to move beyond correlations and toward identification of specific viruses and their precise mechanisms of beneficial or harmful immunomodulation.

  • A major objective of the research presented here is to develop a link between the two approaches by establishing the relationship between the bridge component condition and the capacity and serviceability limits of the structure. A comprehensive review of literature identified methods of predictive modelling of bridge component condition change using different approaches. The review also indicated that a key to the proposed linkage is considering specific degradation mechanisms in predicting bridge deterioration.

  • In this paper residual stress in thick layer composites have been studied. Using numerical analysis, the curing process has been modeled and mechanical properties of a thick layer composite are obtained and the induced residual stress in these composites has been assessed. Unlike thin layer composites, the stress in each layer is not specific and a tensile and compressive stress field can exist simultaneously in thick layer composites.

  • Regulator of G protein signaling (RGS) proteins are gatekeepers regulating the cellular responses induced by G protein-coupled receptor (GPCR)-mediated activation of heterotrimeric G proteins. Specifically, RGS proteins determine the magnitude and duration of GPCR signaling by acting as a GTPase-activating protein for Gα subunits, an activity facilitated by their semiconserved RGS domain. The R7 subfamily of RGS proteins is distinguished by two unique domains, DEP/DHEX and GGL, which mediate membrane targeting and stability of these proteins.

  • This dissertation examines individuals’ actions to improve social outcomes when unrecoverable investments are necessary. Situations involving non-pecuniary and pecuniary investments are considered. In the former, the prerequisite of real effort - a non-pecuniary, unrecoverable investment - is examined when said effort determines an individual’s ability to procure their preferred social outcome.

  • There is an increasing interest in the use of lanthanides in medicine. However, themechanismof their accumulation in cells isnotwell understood.Lanthanidecations are similar to ferric ions with regard to transferrin binding, suggesting transferrin-receptormediated transport is possible; however, this has not yet been confirmed. In order to clarify this mechanism, we investigated the binding of Yb 3+ to apo-transferrin by UV-Vis spectroscopy and stopped-flow spec-trophotometry, and found that Yb 3+ binds to apotransferrin at the specific iron sites in the presence of bicarbonate. ...

  • Ammodytoxins (Atxs) are presynaptically acting snake venom phospholipase A2 (PLA2) toxins the molecular mechanism of whose neurotoxicity is not completely understood. Two chimeric PLA2s were prepared by repla-cing the C-terminal part of a nontoxic venom PLA2 , ammodytin I2, with that of AtxA(K108N). The chimeras were not toxic, but were able to bind strongly to an Atxs-specific neuronal receptor, R25.

  • Dehydroascorbate reductase (DHAR) reduces dehydro-ascorbate (DHA) to ascorbate with glutathione (GSH) as the electron donor. We analyzed the reactionmechanismof spinach chloroplast DHAR, which had a much higher reaction specificity for DHA than animal enzymes, using a recombinant enzyme expressed inEscherichia coli. Kinetic analysis suggested that the reaction proceeded by a bi-uni-uni-uni-ping-pong mechanism, in which binding of DHA to the free, reduced formof the enzyme was followed bybindingofGSH....
