Shikimate kinase was chosen as a convenient representative example of the subclass of a/b proteins with which to examine the mechanism of protein folding. In this paper we report on the refolding of the enzyme after denaturation in urea. As shown by the changes in secondary and tertiary structure monitored by far UV circular dichroism (CD) and fluorescence, respectively, the enzyme was fully unfolded in 4 M urea. From an analysis of the unfolding curve in terms of the two-state model, the stability of the folded state could be estimated as 17 kJÆmol)1.

To examine functions of two small heat shock proteins of Escherichia coli, IbpA and IbpB, we constructed His–IbpA and His–IbpB, in which a polyhistidine tag was fused to the N-terminals. To search for novel regulatory regions, we examined the features of chromatin structure in the rat B29/Ig-b gene and its flanking regions by determining DNase I hypersensitive sites (DHS) in plasmacytoma-derived Y3 cells. Six Y3 cellspecific DHS were detected at )8.6, promoter, +0.7, +4.4, +6.0, and +8.7 kb. The DHS at +4.4, +6.0, and +8.7 kb were present in the intergenic region between B29/Ig-b and growth hormone (GH) genes and were mapped inside conserved sequences in rat and humans.

Alcohol dehydrogenase (ADH) deficiency results in decreased retinol utilization, but it is unclear what physio-logical roles the several known ADHs play in retinoid signaling. Here, Adh1, Adh3,andAdh4null mutant mice have been examined following acute and chronic vitamin A excess. Following an acute dose of retinol (50 mgÆkg )1 ), metabolism of retinol to retinoic acid in liver was reduced 10-fold inAdh1mutants and 3.8-fold inAdh3mutants, but was not significantly reduced inAdh4mutants.
