The binding of Thermomyces lanuginosa lipase and its mutants [TLL(S146A), TLL(W89L), TLL(W117F, W221H, W260H)] to the mixed micelles of cis-parinaric acid/ sodium taurodeoxycholate at pH 5.0 led to the quenching of the intrinsic tryptophan fluorescence emission (300–380 nm) and to a simultaneous increase in the cis-parinaric acid fluorescence emission (380–500 nm). These findings were used to characterize the Thermomyces lanuginosa lipase/cisparinaric acid interactions occurring in the presence of sodium taurodeoxycholate....

The ¯uorescence resonance energy transfer parameter, f, is de®ned as the eciency of the energy transfer normalized by the quantum yield of the donor in the presence of acceptor. It is possible to characterize the ¯exibility of the protein matrix between the appropriate ¯uorescent probes by monitoring the temperature dependence of f. The intermonomer ¯exibility of the Ca-actin and Mg-actin ®laments was characterized by using this method at pH values of 6.5 and 7.4.

Mammalian adenylate cyclases are predicted to possess complex topologies, comprising two cassettes of six trans-membrane-spanningmotifs followedbyacytosolic, catalytic ATP-binding domain. Recent studies have begun to provide insights on the tertiary assembly of these proteins; crystal-lographic analysis has revealed that the two cytosolic domains dimerize to forma catalytic core, whilemore recent biochemical and cell biological analysis shows that the two transmembrane cassettes also associate to facilitate the functional assembly and trackingof the enzyme. ...

The calcium-sensing receptor (CaR) belongs to family C of the G-protein coupled receptor superfamily. The receptor is believed to exist as a homodimer due to covalent and non-covalent interactions between the two amino terminal domains (ATDs). It is well established that agonist binding to family C receptors takes place at the ATD and that this causes theATDdimer to twist. However, very little isknown about the translationof theATDdimer twist intoG-protein coupling to the 7 transmembrane moieties (7TMs) of these receptor dimers....

Human D2Long(D2L)andD2Short(D2S) dopamine receptor isoforms were modified at their N-terminus by the addition of a human immunodeficiency virus (HIV) or a FLAG epitope tag. The receptors were then expressed inSpodop-tera frugiperda 9(Sf9)cells using the baculovirus system, and their oligomerization was investigated by means of co-immunoprecipitation and time-resolved fluorescence resonance energy transfer (FRET).

A new baculovirus-based fluorescence resonance energy transfer (Bv-FRET) assay for measuringmultimerization of cell surface molecules in living cells is described. It has been demonstrated that gonadotropin-releasing hormone recep-tor (GnRH-R) was capable of forming oligomeric com-plexes in the plasma membrane under normal physiological conditions. The mouse gonadotropin-releasing hormone receptor GnRH-R was used to evaluate the efficiency and potential applications of this assay.

A wide range of approaches has been applied to examine the quaternary structure of G protein-coupled receptors, the basis of such protein–protein interactions and how such interactions might modulate the pharmacology and function of these receptors. These include coimmunoprecipitation, var-ious adaptations of resonance energy transfer techniques, functional com-plementation studies and the analysis of ligand-binding data.

Rad51 and disrupted meiotic cDNA1 (Dmc1) are the two eukaryotic DNA recombinases that participate in homology search and strand exchange reactions during homologous recombination mediated DNA repair. Rad51 expresses in both mitotic and meiotic tissues whereas Dmc1 is confined to meiosis.

The tumor suppressor von Hippel–Lindau (VHL) gene product forms a com-plex with elongin B and elongin C, and acts as a recognition subunit of a ubiquitin E3 ligase. Interactions between components in the complex were investigated in living cells by fluorescence resonance energy transfer (FRET)–fluorescence lifetime imaging microscopy (FLIM).

The present study aimed to investigate the role played by the leaflets of the plasma membrane in the uptake of drugs into cells and in their extrusion by P-glycoprotein and multidrug resistance-associated protein 1. Drug accumulation was monitored by fluorescence resonance energy transfer from trimethylammonium-diphenyl-hexatriene (TMA-DPH) located at the outer leaflet to a rhodamine analog.
