Tumour necrosis factor-related apoptosis-inducing ligand (TRAIL) has attractedmuch attention because of its ability to kill tumour cells. In this study, we demonstrated that treatment of QGY-7703 cells with the combination of TRAIL and etoposide resulted in synergistic cytotoxic effects. In dissecting the mechanism underlying this syner-gistic effect, we found that treatment with etoposide alone resulted in the upregulation of Bax, while the level of trun-catedBid (tBid)wasunchanged.

Endotoxin-neutralizingprotein (ENP) of the horseshoe crab isoneof themost potent neutralizersof endotoxins [bacterial lipopolysaccharide (LPS)]. Here, we report on the inter-action of LPS with recombinant ENP using a variety of physical and biological techniques. In biological assays (Limulusamebocyte lysate and tumour necrosis factor-a induction in human mononuclear cells), ENP causes a strong reduction of the immunostimulatory ability of LPS in a dose-dependent manner.

Tumour cells are often sensitized by interferons to the effects of tumour necrosis factor-a-related apoptosis-inducing ligand (TRAIL). We have demonstrated previously that TRAIL has an inhibitory effect on protein synthesis [Jeffrey IW, Bushell M, Tilleray VJ, Morley S & Clemens MJ (2002) Cancer Res62, 2272–2280] and we have therefore examined the consequences of prior interferon-atreatment for the sensitivity of transla-tion to inhibition by TRAIL.

Circulating tumour necrosis factor-a(TNFa) levels, which are elevated in obesity-associated insulin resistance and diabetes, inhibit insulin signalling at several points in the signalling cascade. The liver is critical in maintaining cir-culating glucose levels and, in a preliminary investigation using the human hepatoma (HepG2) cell line in this study, we demonstrated the role of TNFa in the regulation of this phenomenon and determined the underlying molecular mechanisms.

A novel geneZC3H12A, encoding MCP-1-induced protein 1 (MCPIP), was recently identified in human peripheral blood monocytes treated with monocyte chemotactic protein 1 (MCP-1) and in human monocyte-derived macrophages stimulated with interleukin (IL)-1b. These experiments revealed that the gene undergoes rapid and potent transcription induction upon stimulation with proinflammatory molecules, such as MCP-1, IL-1b, tumour necrosis factor aand lipopolysaccharide.
