This study aims to determine the various strains of yeast and filamentous fungi in traditional alcohol yeast cake from Hai Hau district, Nam Dinh province. By traditional laboratory techniques such as isolation on YPD agar, observation of colony morphology as well as cell characteristics by microscopy, combined with molecular biology method to sequence ITS region 03 strains of yeast Saccharomyces cerevisiae CN1, CN2, and NM3 were identified: 01 pseudo-strain yeast Saccharomycopsis fibuligera NM02 and 01 strain of filamentous fungi Rhizopus microsporus NS04.

In this study, the fruiting bodies of the X. nigripes were collected to isolate, propagate in the liquid medium, and then cultured to induce the fruiting body formation in the rice medium. The results showed that the X. nigripes was successfully isolated on PDAY (Potato Dextrose Agar Yeast Extract) medium, and the mycelium and spore of the X. nigripes had unique characteristics of the species.

Pyruvate decarboxylase (EC 4.1.1.1) was isolated and purified from the yeast Kluyveromyces lactis. The properties of this enzyme relating to the native oligomeric state, the subunit size, the nucleotide sequence of the coding gene(s), the catalytic activity, and protein fluorescence as well as circular dichroism are very similar to those of the well characterized pyruvate decarboxylase species from yeast.

An oxidosqualene cyclase cDNA, LcIMS1, was isolated from cultured cells of Luffa cylindrica Roem. by heterologous hybridization with cDNA of Glycyrrhiza glabra b-amyrin synthase. Expression of LcIMS1 in yeast lacking endogenous oxidosqualene cyclase activity resulted in the accumulation of isomultiflorenol, a triterpene. This is consistent with LcIMS1 encoding isomultiflorenol synthase, an oxidosqualene cyclase involved in bryonolic acid biosynthesis in cultured Luffa cells.

The oxidant function of pro-apoptotic protein Bax was investigated through heterologous expression in yeast. Direct measurements of fatty acid content show that Bax-expression induces oxidation of mitochondrial lipids. This effect is prevented by the coexpression of Bcl-xL. The oxi-dation actually could be followed on isolated mitochondria as respiration-induced peroxidation of polyunsaturatedcis-parinaric acid and on whole cells as the increase in the amount of thiobarbituric acid-reactiveproducts. ...

Mitochondrial ribosomal proteins (mrps) of the budding yeast, Saccharomyces cerevisiae, have been extensively characterized genetically and biochemically. However, the list of thegenes encoding individualmrps is still not complete and quite a fewof the mrps are only predicted from their similarity to bacterial ribosomal proteins. We have con-structed a yeast strain in which one of the small subunit proteins, termed Mrp4, was tagged with S-peptide and used for affinity purification of mitochondrial ribosome. ...

In eukaryotes, pre-rRNA processing depends on a large number of non-ribosomal trans-acting factors that form large and intriguingly organized complexes. A novel nucleolar protein, Nop53p, was isolated by using Nop17p as bait in the yeast two-hybrid system. Nop53p also interacts with a second nucleolar protein, Nip7p. A carbon source-conditional strain with the NOP53coding sequence under the control of theGAL1promoter did not grow in glucose-containing medium, showing the phenotype of an essential gene. ...

We isolated two novel 14-3-3 binding proteins using 14-3-3fas bait in a yeast two-hybrid screen of a human brain cDNA library. One of these encoded the C-terminus of a neural specific armadillo-repeat protein, d-catenin (neural plakophilin-related arm-repeat protein or neurojungin). d-Catenin from brain lysates was retained on a 14-3-3 affinity column.

Whereas the majority of disease-related mitochondrial DNA mutations exhibit significant biochemical and clinical heterogeneity, mutations within the mitochondrially encoded human cytochrome b gene (MTCYB) are almost exclusively associated with isolated complex III deficiency in muscle and a clinical presentation involving exercise intolerance.

Saccharomyces cerevisiaepossesses two glycogenin isoforms (designated as Glg1p and Glg2p) that both contain a con-served tyrosine residue, Tyr232. However, Glg2p possesses an additional tyrosine residue, Tyr230 and therefore two potential autoglucosylation sites. Glucosylation of Glg2p was studied using both matrix-assisted laser desorption ionization and electrospray quadrupole time of flight mass spectrometry. Glg2p, carrying a C-terminal (His6) tag, was produced inEscherichia coliand purified.

During a search for cDNAs encoding plant sterol acyl-transferases, we isolated four full-length cDNAs fromAra-bidopsis thaliana that encode proteins with substantial identity with animal lecithin : cholesterol acyltransferases (LCATs). The expressionof one of these cDNAs,AtLCAT3 (At3g03310), invarious yeast strains resulted in the doubling of the triacylglycerol content.

The cytochromebc1 complex of the yeastSaccharomyces cerevisiae is composed of 10 different subunits that are assembledas a symmetrical dimer in the innermitochondrial membrane. Three of the subunits contain redox centers and participate in catalysis, whereas little is known about the function of the seven supernumerary subunits.

NEDD8 is a ubiquitin-like protein that controls vital bio-logical events through its conjugation to target proteins. Previously, we identifieda negative regulator oftheNEDD8 conjugation system, NEDD8 ultimate buster-1 (NUB1), that recruits NEDD8 and its conjugates to the proteasome for degradation. Recently, we performed yeast two-hybrid screening with NUB1 as bait and isolated a ubiquitin pre-cursor UbC1 that is composed ofnine tandem repeats of a ubiquitin unit through a-peptide bonds.

Aquaporins andaquaglyceroporinsmediate the transport of waterand solutesacrossbiologicalmembranes.Saccharo-myces cerevisiaeFps1 is an aquaglyceroporin that mediates controlled glycerol export during osmoregulation. The transport function of Fps1 is rapidly regulated by osmotic changes in an apparently unique way and distinct regions within the longN-andC-terminal extensions are needed for this regulation.

Type II NADH dehydrogenases (NDH-2) are monomeric enzymes that cat-alyse quinone reduction and allow electrons to enter the respiratory chain in different organisms including higher plant mitochondria, bacteria and yeasts. In this study, anAgrobacterium tumefaciensgene encoding a puta-tive alternative NADH dehydrogenase (AtuNDH-2) was isolated and expressed in Escherichia coli as a (His)6-tagged protein.

The EVI1 transcriptional repressor is critical to the normal development of a variety of tissues and participates in the progression of acute myeloid leu-kaemias. The repressor domain (Rp) was used to screen an adult human kidney yeast two-hybrid library and a novel binding partner designated ubiquitously expressed transcript (UXT) was isolated.
